In this project we aim to investigate the molecular signatures of extracellular matrix (ECM) components during progression of hepatic fibrosis, with a special interest in the hepatic fibrosis resulting from Non-Alcoholic Fatty Liver Disease (NAFLD), where the increasing fat and/or fibrosis is not caused by alcohol abuse. This include two separate origins leading to similar down-stream events. One compromises excess fat deposition in the liver leading to fibrosis and potentially a hepatocellular carcinoma (HCC) and the other Non-Alcoholic steatohepatitis (NASH) originating from enhanced inflammation of the liver leading to similar symptoms as in the Fatty Liver case. NAFLD is linked with the occurrence of obesity and is the most common liver disease in the US. Recent results demonstrate the existence of molecular signatures of ECM components during progression of hepatic fibrosis. Experiments in animal models point to an intimate link between fibrogenesis and tissue stiffness and provide evidence that liver fibrosis is caused by altered biophysical properties of the ECM, such as deposition of fat and crosslinking of collagen fibres. This hints toward a relation between biochemical- and mechanical changes of ECM components, that mutually influence each other by means of molecular signalling. Therefore, we aim to gain new insights into the role of the ECM in hepatic fibrosis, and exploit the potential of non-invasive mechanical imaging by MR elastography (MRE) combined by biochemical assays, such as proteomics, glycosaminoglycan (GAG) analysis, histology and Liver fat fraction analysis to unravel the underlying mechanisms of this disease.
