I developed methods and tools that allowed to characterize the fungal microbiome and polymicrobial interactions in the cystic fibrosis lung. I am also part of a multi-group project that is developing a rapid identification tool for bacterial and fungal infections in the cystic fibrosis lung based on the NanoString technology. Additional, I was the lead scientist of a multi-institutional project that aims to analyze the phosphoproteome of the human fungal pathogen Candida albicans.

Postdoctoral fellow

I was working on understanding how A. fumigatus is able to survive in the hypoxic conditions encountered during fungal pathogenesis. At sites of infection in the lung, significant influxes of host immune effector cells as well as tissue damage create a microenvironment devoid of oxygen in which the fungus must survive. I was using microarray analysis to identify genes involved in the fungal response to hypoxia, and in addition I was working on a specific gene, srbA, that is involved in sensing oxygen and allowing the fungus to grow in hypoxic conditions.

I analyzed the influence of O-glycosylation on the virulence of C. neoformans by characterizing the protein mannosyltransferase (PMT) gene family. This gene family, which consists of three members, PMT1, PMT2, and PMT4, encodes enzymes that catalyze the initial step of protein O-glycosylation. Effects of deleting PMT-genes on virulence of C. neoformans were determined by routine in vitro and in vivo assays analyzing specific virulence factors and infectivity of the pathogen, respectively.
